淋巴结(lymph nodes, LNs)成纤维网状细胞(fibroblastic reticular cells, FRCs)是LNs免疫微环境中的关键基质细胞。FRCs通过分泌趋化因子和细胞因子、构建细胞外基质网络,参与调控免疫细胞的迁移、存活及功能维持,在维持LNs结构完整性和免疫稳态中发挥重要作用。近年来,随着单细胞测序和空间转录组等技术的发展,研究进一步揭示了FRCs的功能异质性及其在人类免疫缺陷病毒(human immunodeficiency virus, HIV)感染中的重要作用。HIV感染可诱发LNs纤维化及FRCs功能紊乱。同时,HIV感染与滤泡树突状细胞间存在协同作用,有助于病毒储存库的形成与维持,并成为抗逆转录病毒治疗后病毒反弹的重要原因。本文系统综述了FRCs的生物学特性、HIV感染对其功能的影响以及靶向FRCs的治疗策略,以期为理解HIV相关免疫病理机制和开发新型治疗靶点提供理论依据。
Abstract:Lymph node fibroblastic reticular cells(FRCs) are key stromal components of the lymph nodes(LNs) immune microenvironment. By secreting chemokines and cytokines and constructing the extracellular matrix network, FRCs regulate migration, survival, and functional maintenance of immune cell, thereby playing a crucial role in preserving LN structural integrity and immune homeostasis. In recent years, advances in technologies such as single-cell sequencing and spatial transcriptomics have further revealed the functional heterogeneity of FRCs and their important role in human immunodeficiency virus(HIV) infection. HIV infection induces LNs fibrosis and dysfunction of FRCs. Moreover, the synergistic interaction between HIV and follicular dendritic cells facilitates the establishment and persistence of viral reservoirs, representing a major cause of viral rebound after antiretroviral therapy. This review systematically summarizes the biological characteristics of FRCs, the impact of HIV infection on their functions and the therapeutic strategies targeting FRCs, with the aim of providing a theoretical basis for understanding HIV-related immunopathogenesis and developing novel therapeutic targets.
